DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The subchronic toxicity of diflorasone diacetate (I) [33564-31-7], a new topical anti-inflammatory agent, was studied in rats of both sexes. The drug was administered s.c. at daily doses of 0.0045, 0.045, 0.45, 2.25 and 11.25 mg/kg for 13 wk. In rats given diflorasone diacetate at 30.45 mg/kg, depression in body wt. gain, decrease of peripheral leukocyte (lymphocyte) count, atrophy of spleen, thymus and adrenal glands and infiltration of fat droplets in hepatic cells were obsd. All these changes disappeared or were reduced 10 wk after cessation of administration.

(fluocinonide, betamethasone 17-valerate, and fluocinolone acetonide) when the compds. were dissolved in 95% alc. and applied in vasoconstrictor assays in healthy volunteers. On the basis of addnl. vasoconstrictor assay results, a 0.05% concn. of I in a cream vehicle contg. 15% propylene glycol was developed for therapeutic evaluation. In a double-blind comparison in 384 patients with dermatoses, 0.05% I cream was as effective as 0.05% fluocinonide cream in the therapy of lesions of psoriasis or atopic/neurodermatitis.
