Conjugated metabolites are split by enzyme hydrolysis. After addn. of an internal std., I is extd. at acid pH into petroleum ether, transferred to an alk. aq. soln., and back-extd. into petroleum ether after acidification. Both compds. are acetylated with acetic anhydride in the presence of pyridine. The acetyl derivs. are detd. by GLC using a 63-Ni-electron-capture detector. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) A gas chromatog. method is presented for the detn. of Irgasan DP 300 (2-hydroxy-2',4,4'-trichlorodiphenyl ether)(I) [3380-34-5] in blood, urine, bile, etc. The method is capable of detecting 10 .mu.g I/L. In comparison with enzymic hydrolysis, acid hydrolysis of the conjugate is an improvement with respect to speed and effectiveness. Radiochem. checking shows reproducible recoveries, 95-105% for free I and 80-95% for Irgasan DP 300 glucuronide [63156-12-7].
