DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The dopaminergic actions of various hallucinogenic drugs administered i.p. were assessed by examg. their effects on turning behavior in rats with unilateral 6-hydroxydopamine lesions of the nigrostriatal pathway. LSD bitartrate (I bitartrate) [113-41-7] (0.1 and 0.2 mg/kg) produced strong contralateral turning, indicating that it is a potent dopamine [51-61-6] receptor agonist, whereas 2-bromo-d-lysergic acid diethylamide bitartrate [4004-43-7] (5 mg/kg), a nonhallucinogenic congener of I, was a weak dopamine receptor agonist. 2,5-Dimethoxy-4-methylamphetamine-HCl [15589-00-1] (2 and 5 mg/kg) and mescaline [54-04-6] (50 and 100 mg/kg) produced ipsilateral turning, indicating that these compds. have a moderate dopamine-releasing action. N,N-dimethyltryptamine-HCl [2826-93-9] (10 and 20 mg/kg) and 5-methoxy-N,N-dimethyltryptamine-HCl [2427-79-4] (0.75 and 1.25 mg/kg) produced weak ipsilateral turning, which was not significantly different from that produced by the nonhallucinogenic compds. tryptamine-HCl [343-94-2] (40 mg/kg) or scopolamine-HBr [114-49-8] (0.25 mg/kg). Psilocin (1-20 mg/kg) produced no turning in either direction. These data, in conjunction with previous studies, indicate that whereas inactivation of the brain serotonin system may be a necessary and sufficient condition for hallucinogenesis, the ability to activate dopamine receptors may be an important factor in detg. the potency of hallucinogens.
