DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The F344 rat liver microsomes, in the presence of an NADPH-generating system, can metabolize methylazoxymethanol (MAM) [590-96-5], a colon and liver carcinogen, to MeOH [67-56-1] and HCO2H [64-18-6]. This is in contrast to the spontaneous decompn. of MAM which yields MeOH and HCHO. The metab. of MAM by rat liver microsomes is sensitive to inhibition by CO as well as to inhibition by 3-methylpyrazole (3-MeP) [1453-58-3] and 4-iodopyrazole (4-IP) [3469-69-0], the latter being more potent than 3-MeP. Pretreatment of rats with 4-IP decreased the level of MAM acetate [592-62-1]-induced DNA methylation in both the liver and the colon mucosa. In contrast, pretreatment with 3-MeP decreased MAM acetate-induced liver DNA methylation, but increased DNA methylation in the colon mucosa. This differential effect of the 2 compds. on DNA methylation in the liver and the colon suggests that different enzymes are responsible for activation of the carcinogen in the 2 organs. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Four daily doses of pyrazole [288-13-1] (50 mg/kg) decreased rat brain noradrenaline (I) [51-41-2] by >20% when detd. 24 h after the final injection. Neither 4-methylpyrazole [7554-65-6] (10-50 mg/kg) nor 4-iodopyrazole [3469-69-0] (10-50 mg/kg) had any effect. In mice treated similarly, pyrazole (50-400 mg/kg) caused a dose-dependent decrease in brain I. Neither 4-methylpyrazole, 4-bromopyrazole [2075-45-8], nor 4-iodopyrazole caused any significant change in the levels. However, if the brain I levels were examd. 6 h after a single dose, then in addn. to pyrazole, 4-methylpyrazole showed a dose-dependent ability to lower brain I. 4-Bromopyrazole and 4-iodopyrazole, given acutely, caused a dose-dependent decrease in rectal temp. and exploratory behavior. 4-Methylpyrazole in high doses (200-400 mg/kg) showed similar properties but they did not correlate with the decrease in brain I. Pyrazole, after acute treatment, showed little ability to change rectal temp. or exploratory behavior. Apparently, the I-depleting effect of pyrazole is not related to inhibition of alc. dehydrogenase, since other 4-substituted pyrazoles which are more potent inhibitors of the enzyme have little or no effect on brain NA levels. .
