In a sep. expt., the melatonin concn. in the media was also periodically measured by RIAto det. the viability of the organ culture over 4 days. The melatonin level on day 2 (2321 pg/gland) was higher than on day 3 (1542 pg/gland) or day 4 (805 pg/gland). The results verify the viability of the hamster pineal organ culture and show that the gland responds to NE in vitro. .

The improvements consisted of using MeCN instead of MeOH in the eluant, a slightly higher pH (5.1), and a more efficient reverse-phase column (Ultrasphere C18). A complete anal. required less than the equiv. of 2 rat pineal glands. Samples were obtained at 8 time points. A reciprocal physiol. relation was found between oxidn. and N-acetylation products of 5-HT. 5-HT and the oxidn. products 5-HIAA, 5-hydroxytryptophol, 5-methoxytryptophol, and 5-methoxyindoleacetic acid decreased at night, when the N-acetylation products N-acetylserotonin and melatonin increased. These observations are consistent with the hypothesis that circadian changes in the N-acetylation of 5-HT by indoleamine N-acetyltransferase [9027-33-2] is the major factor controlling circadian changes in the amts. of 5-HT, and 5-HT oxidn. and N-acetylation products in the rat pineal gland. .
