DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) Following infection with B. fragilis in mice there was a fall in viable bacterial nos. over the first 24 h, followed by a recovery phase and subsequent regrowth of the organism in the groin tissue, resulting in abscess formation. The most effective therapy usually began shortly after infection. There was little correlation between groin counts and palpable abscesses; assessment of therapy was best achieved by bacterial groin counts. In this infection model relatively high doses of b-lactam antibiotics were required for effective treatment. This was not due to inadequate penetration at the site of infection as high levels were measurable in the pus, but it was more likely due to inactivation by the B. fragilis b-lactamase [9073-60-3]. Therapy with the b-lactamase inhibitor clavulanic acid [58001-44-8] overcame the problem of b-lactamase instability of ticarcillin [34787-01-4] and amoxicillin [26787-78-0], resulting in highly effective therapy with both ticarcillin and amoxicillin in the presence of clavulanic acid. .

These strains were also susceptible to other ureidopenicillins. Similar selections of transformants with carbenicillin [4697-36-3], ampicillin [69-53-4], or ticarcillin [34787-01-4] required high concns. of antibiotics and yielded backgrounds of spontaneous resistant mutants. .
