DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The LD50 values of NK 19 (I) [3571-88-8] were 46.9 and 50.5 mg/kg, resp., for male and female mice via the i.p. route. The values were 1539 and 1571 mg/kg, resp., for male and female rats after oral administration. Poisoning symptoms included hair bristling, short breathing, convulsion, liver and kidney parenchymatous degeneration and others. In subacute and chronic expts., I (at 1 and 5 mg/kg for 5 weeks or at 0.2, 1, and 5 mg/kg for 15 weeks) administered i.p. to rats did not cause changes in the body wt., food consumption, and pathohistol. parameters.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The effect of cyanine and aminovinyl dyes on the compd. 48/80-induced degranulation and histamine [51-45-6] release by isolated rat mast cells was studied. Of 12 cyanine dyes tested, NK 76 (I; n = 1) [905-97-5], NK 136 (I; n = 2) [514-73-8], NK 19 (II; R = C7H15) [3571-88-8], NK 2251 [53213-94-8], NK 266 [37187-89-6], and NK 2252 [62008-59-7] inhibited the compd. 48/80-induced degranulation, whereas NK 88 (I; n = 0) [2197-01-5], NK 2 (II; R = Me) [37187-85-2], NK 2011 [62008-57-5], NK 1160 [62008-58-6], NK 1529 [62008-60-0] and NK 4 [4846-34-8] had no effect. All aminovinyl dyes (NK 343 [1463-95-2] and NK 355 [21034-17-3]) tested were without effect. NK 2251 also inhibited compd. 48/80- or ATP [56-65-5]-induced histamine release but had no effect on ATP-induced cell swelling. The mechanism of histamine release is discussed.
