DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The principal excretion products derived from radiolabeled N,N-dimethylaminoisopropanol (Dip) [108-16-7] and p-acetamidobenzoic acid (PAcBA) [556-08-1] components of inosiplex (Isoprinosine) [36703-88-5] were identified and quantified in urine following single i.v. and oral administration of the drug in rhesus monkeys. The major metabolite derived from [3H]PAcBA was identified as PAcBA-O-acylglucuronide [34220-56-9] by 1) pos. naphthoresorcinol reaction for glucuronic acid and 2) hydrolysis of the metabolite to PAcBA and glucuronic acid, using either dil. 1984-38-9 and 108-16-7 which are cas registry numbers are also used here. base (but not acid) or b-glucuronidase. This metabolite accounted for 50% of the administered dose in orally dosed animals and 31% in i.v. dosed animals. A minor metabolite, which constituted ~5% of the excreted 3H from either i.v. or orally dosed animals, was identified as the hippuric acid conjugate of PAcBA [1984-38-9] by cochromatog. with a com. std. A single metabolite derived from [14C]Dip was identified as Dip-N-oxide [27607-29-0] by cochromatog. with synthetic material in several chromatog. systems; this metabolite accounted for 17-18% of the administered 14C in either the i.v. or orally dosed animals. .
