Thus, Na xylan sulfate (I) [37319-17-8] (SP 54) was converted by ion exchange to a pyridine salt which was heated in DMSO to give desulfated xylan, the H2O-insol. fraction of which (29.4%) was sulfated with ClSO3H in pyridine-DMF and converted by NaOH to I with no.-av. mol. wt. 7690, degree of sulfation 1.72, uronic acid content 4.98%, and pentose content 30.4%. This I had anticoagulant activity (activated cephalin time) 129 and 298% prolongation at 2.5 and 5 mg/kg, resp., and antithrombotic activity (anti-Xa) 57% prolongation, compared to 104, 200, and 25, resp., for SP 54, and 1178, incoagulable, and 506, resp., for heparin.

Sulfate polyanions prolong the incubation period of scrapie-infected hamsters. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effect of the org. sulfonated polyanions, pentosan sulfate (SP54), dextran sulfate 500 (DS500) and suramin, have been tested on golden Syrian hamsters infected with the 263K strain of scrapie by the i.p. or the intracerebral route. SP54 had the greatest effect in prolonging the incubation period of the disease when administered within 2 h of the i.p. inoculum. 145-63-1 and 37319-17-8 are cas registry numbers of chemicals which are used as reagents here. The same amt. of SP54 given 24 h after scrapie inoculation had a potent effect in some animals and no effect in others. This result suggests that SP54 inhibits the uptake of the scrapie agent into the nerve endings and/or carrier cells at the site of the inoculum, i.e. the peritoneum, and that this event occurs in about 24 h. DS500 had a similar although less potent effect (22.4 days delay during the incubation period) than SP54 (54.4 days) when administered within 2 h of scrapie injection by the i.p. route, and suramin had only a minimal effect (10 days). This study suggests that treatment of scrapie and related spongiform encephalopathies of animals and man is possible only before the agent has reached the clin. target areas of the brain. .
