DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Section cross-reference(s): 32 To obtain specific antisera for the enzyme immunoassay of dexamethasone [50-02-2], dexamethasone-21-hemisuccinate (I) [3800-86-0], dexamethasone-6-carboxymethyl thioether (II) [88378-31-8], and dexamethasone-3-(O-carboxymethyl)oxime (III) [88378-32-9] were synthesized. Each compd. was coupled with bovine serum albumin (BSA) by the mixed anhydride or carbodiimide method, and the complex was used to raise antisera in rabbits. Alk. phosphatase was conjugated with I, II, and III using water-sol. carbodiimide. Bound and free dexamethasones were sepd. by the double antibody method. The specificity of each antiserum was assessed by detg. cross-reactivity of other corticoids by an enzyme immunoassay. Anti-II-BSA and anti-III-BSA antisera had a higher specificity than did anti-I-BSA antisera. Anti-II-BSA and anti-III-BSA antisera were purified by affinity chromatog. packed with betamethasone-3-(O-carboxymethyl)oxime [61600-49-5]-, II-, and III-Sepharose 4B, resp. The percent cross-reactivities against betamethasone, a C-16 epimer of dexamethasone, of purified anti-II-BSA and anti-III-BSA antisera decreased remarkably. Using these purified antisera, it was possible to det. the amt. of 0.2-20 ng/assay tube of dexamethasone, in the presence of 100-fold betamethasone. .
