DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Diphenoxylate-HCl (I-HCl) [3810-80-8] and difenoxine-HCl (II-HCl) [35607-36-4] (which were structurally related to the narcotic analgesic meperidine) possessed both antidiarrheal and analgesic effects that could not be sepd. by any of the tests used to measure these properties. In contrast, loperamide-HCl (III-HCl) [34552-83-5] (which was unrelated to any narcotics) showed antidiarrheal activity that was not assocd. with analgesic effects in all tests. In order to avoid potential addiction problems, care must be taken in the development of antidiarrheal agents related to mols. with central analgesic effects.
