DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Section cross-reference(s): 1 Resoln. of oxyphenonium [14214-84-7] and some quaternary atropine analogs was attempted with a chiral-HPLC system, contg. d-camphorsulfonic acid as a mobile phase component or with chiral stationary phases. Neither approach was successful, due to the relatively long distance between the chiral center and the quaternized nitrogen. Therefore resoln. of the optical antipodes of the acid parts of the cholinergics (easily obtained by hydrolysis) was attempted. An HPLC system having b-cyclodextrin [7585-39-9] as the chiral stationary phase was chosen. Using aq. mobile phases, cyclohexylphenylglycolic acid [20585-34-6] (the acid moiety in oxyphenonium), cyclohexylphenylacetic acid [3894-09-5] and mandelic acid [90-64-2] (e.g. in homatropine) were resolved. However, tropic acid [529-64-6] could not be sepd. into enantiomers with this system. Consideration is given to the affect of temp. and sample concn. on selectivity and resoln. and to difficulties in quant. anal. .
