DOCUMENT TYPE: Conference CA Section: 1 (Pharmacodynamics) In isolated frog heart prepns., the pyrimidines, metacil (I) [56-04-2], hydroxymetacil [35463-39-9], superacil [3977-29-5] isocytosine [108-53-2], pentoxyl [147-61-5], and uracil [66-22-8], increased the amplitude of contractions. The pyrimidines also enhanced the effect of strophanthin on isolated frog heart contraction. Metacil and hydroxymetacil increased the amplitude of cardiac contraction in rabbits with exptl. infarcts 3-fold and in rabbits with exptl. myocardites .apprx.1.5 fold.

Pentoxyl [147-61-5] had no inotropic effects and none of the pyrimidine derivs. had chronotropic effects. In rabbits with exptl. myocardial infarction, I and hydroxymetacil increased amplitude of cardiac contraction 305 and 298%, resp. In rabbits with exptl. myocarditis hydroxymetacil had the greatest cardiotonic effect increasing cardiac contraction 156.6%.
