Effect of a myo-inositol antagonist, 2-O,C-methylene-myo-inositol, on the metabolism of myo-inositol-2-3H and D-glucose-1-14C in Lilium longiflorum pollen. DOCUMENT TYPE: Journal CA Section: 5 (Agrochemicals) Section cross-reference(s): 11 2-O,C-methylene-myo-inositol (I) [4068-87-5], a myo-inositol (MI) [87-89-8] antagonist, inhibited germination and tube elongation of pollen from lily (Lilium longiflorum). The presence of 5 mM I in Dickinson's pentaerythritol medium partially blocked germination. The tubes produced were short and failed to elongate. In the presence of MI, I's toxic effect was blocked. As little as 0.56 mM MI maintained normal germination in the presence of 43 mM I, and pollen tubes continued to elongate for 2 to 3 h. Eventually, the toxic action of I prevented further growth. I did not inhibit UDP-D-glucose dehydrogenase from lily pollen. Uptake of MI-2-3H and incorporation of tritium into galacturonic acid and pentose units of tube wall pectin [9000-69-5] was blocked by I. The site of this inhibition was undertermined. Uptake of 14C-labeled D-glucose [50-99-7] and incorporation of 14C into 70% ethyl alc.-insol. polysaccharides of germinated pollen was not blocked by I, but distribution of label into polysaccharide product was altered. In I-treated pollen, very little 14C was found in uronic acid or pentose units. At 30 mM I, about 2-3 of the C flow from D-glucose to these pectic components was interrupted. I also altered D-glucose metab. in the 70% ethyl alc.-sol. fraction, but the compd. involved must still be identified. These results offer fresh evidence of an intermediary role for MI during UDP-D-glucuronic acid [2616-64-0] biosynthesis in germinated pollen.
