DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Primary cocultures of Sertoli and germ cells were prepd. from the testis of rats and were used for studying testicular damage produced by di(2-ethylhexyl) phthalate [117-81-7] and its metabolites [mono(2-ethylhexyl) phthalate [4376-20-9]; mono(5-carboxy-2-ethylpentyl) phthalate [40809-41-4]; mono(2-ethyl-5-oxohexyl) phthalate [40321-98-0]; and mono(2-ethyl-5-hydroxyhexyl) phthalate [40321-99-1]] and by AF1312/TS [50264-86-3]. Although enhancement of germ cell detachment was a sensitive index of testicular toxicity for phthalate esters and AF1312/TS, this was not the case for other chems. that produce differing patterns of testicular injury in vivo, e.g., methoxyacetic acid [625-45-6] caused several changes but propoxyacetic acid [54497-00-6] and butoxyacetic acid [2516-93-0] had no effect.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The absorption, distribution, and metabolic excretion of mono-2-ethylhexyl phthalate (I) [4376-20-9] were studied in the rat. This compd. was readily adsorbed from the gastrointestinal tract. Following i.v. administration, I was rapidly distributed in all tissues, with the highest levels occurring in the liver, kidney, and urinary bladder. Excretion of I was rapid, ~80% of the dose was eliminated 24 h after oral administration, 72% in the urine and 8% in feces. I was extensively metabolized after oral administration, and the major urinary metabolites were identified as the alc. [40321-99-1], ketone [40321-98-0], and acid [40809-41-4] resulting from the side-chain oxidn. of I. A trace of o-phthalic acid [88-99-3] was also identified.
