DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The EKG cardiovascular dynamics and cardiac contractility was examd. in 7 dogs following administration of DIV 154 (N-benzo[b]-furan-2-ylcarbonyl-N'-benzylpiperazine hydrochloride)(I) [41716-84-1] (0.3, 1.0, 3.0 mg/kg i.v.) and imipramine (0.3, 1.0, 3.0 mg/kg i.v.). The mean arterial blood pressure remained unaltered under the influence of I. However, imipramine, 3 mg/kg i.v. induced a marked decrease in blood pressure which was coupled with a persistent redn. in the total peripheral resistance. Although imipramine (1 mg/kg i.v.) elicited a pronounced chronotropic activity, I (1 and 3 mg/kg i.v.) caused a heart rate increment of a mere few beats. The cardiac output and stroke vol. indices showed an initial increase after the application of 0.3 and 1 mg/kg i.v. imipramine as was the case with I, thereafter a moderate continuous diminution occurred which was more pronounced under the influence of imipramine. The cardiac work was ony slightly diminished by I in the dosage range of 0.3 to 3 mg/kg i.v., whereas 3 mg/kg i.v. imipramine elicited a marked redn. in this parameter. The left ventricular contractility was scarcely altered by 0.3-3 mg/kg I i.v. in comparison with imipramine (3 mg/kg i.v.), which produced a decrease in the dp/dt value and a moderate diminution in the quotient dp/dt/(p-edp)+c. No change appeared in the EKG pattern following the infusion of 0.Several reagents such as 41716-84-1 is used here.3 mg/kg/min I i.v. until a total dosage of 77.3 mg/kg was attained in contrast to the observable morphol. alterations in the EKG which began to appear following total dosages of 18-23 mg/kg imipramine i.v. .
