DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Pregnant rats were orally treated with BD 40A (formoterol fumarate)(I) [43229-80-7] or hexoprenaline as the pos. control agent from day 7 to day 17 of gestation at the doses of 0.2, 6.0 and 60.0 mg/kg of BD 40A or 6.0 and 60.0 mg/kg of hexoprenaline. No clin. symptoms were obsd. in dams. Though food and water intakes of dams showed transient decreases after treatment, body wt. was higher than the controls. Necropsy done at cesarian sectioning showed, heart wt. of dams were increased in the higher dosage groups of BD 40A and hexoprenaline; but these changes were recovered at the weaning time. In the fetuses a slight retardation of growth was obsd. with 6.0 and 60.0 mg/kg of BD 40A or 6 mg/kg of hexoprenaline. BD 40A caused no increase of external or visceral abnormalities, but wavy ribs were obsd. at 60.0 mg/kg of BD 40A. However this wavy ribs was repaired in the progress of ossification with growth of the pups and it was not obsd. in the weanlings.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) In rats orally treated with BD 40A (I) [43229-80-7] during the peri- and postnatal periods, increased body wt. gains were obsd. in the dams treated with 6.0 or 30.0 mg I/kg during the lactation period, but food and water intake decreased. Increases in the no. of stillborns was obsd. at 6.0 and 30.0 mg I/kg, and the pups born in these dose groups showed less viability and suppression of body wt. gain after birth. The results of the cross fostering study suggested that these low viabilities of pups were caused by I treatment of the dams during gestation and treatment after parturition had no effect. Also, insufficient lactation might have occurred in the I treated dams which may have caused the suppression of body wt. gains of these pups. However, no other effect of BD 40A was obsd. in the functional development or reproductive capacities of newborns (F1) as well as the next generations (F2).
