DOCUMENT TYPE: Conference CA Section: 1 (Pharmacodynamics) Both the vol. of filtrate and the no. of erythrocytes/mm3 which passed per unit time from heparinized human blood through an 8-.mu.m capillary system were reduced by making the blood hyperosmolar with NaCl. This effect was partially prevented by prior incubation of the blood (2.5 h, 37.degree.) with xanthinol nicotinate (I) [437-74-1] (70 mg/100 mL). The total no. of erythrocytes passing through the capillaries in the I-treated sample was >2-fold that in the hyperosmolar control. The viscosity of neither the whole blood nor the plasma was altered by I, which probably increased erythrocyte flow by raising cell ATP concn.

DOCUMENT TYPE: Conference CA Section: 1 (Pharmacodynamics) During perfusion of the isolated dog pancreas with blood-dextran, glucose [50-99-7] was consumed and the concns. of both lactate [50-21-5] and pyruvate [127-17-3] in the perfusate rose, the former to the greater extent. In the presence of xanthinol nicotinate (I) [437-74-1] the rate of glucose consumption was increased 1.6-fold and the lactate-to-pyruvate ratio was shifted in favor of pyruvate. The rise in pyruvate was greater with an alk. perfusion medium than with an acidic one, whereas the effect of I on lactate was independent of pH.
