DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) In cultured human skin-derived fibroblast monolayers, steroids which exhibit glucocorticoid and antiinflammatory activity stimulated the uptake of the amino acid analog .alpha.-aminoisobutyric acid (AIB). Stimulation of AIB uptake was max.In this article, certain chemicals are used. Some of their cas registry numbers are 124-94-7 and 50-02-2 after 5 h of exposure to steroid, persisted .ltoreq.24 h in the presence of steroid, and was blocked by cycloheximide. Since the rate of AIB efflux from monolayers was also accelerated in the presence of steroid, the increase in the rate of AIB uptake is attributable to a greater stimulation of the rate of AIB influx. In the presence of steroid, the apparent Km of uptake of AIB was decreased without an appreciable effect on the apparent Vmax of uptake. Steroid analogs lacking an 11.beta. substituent neither stimulated the uptake of AIB not blocked the action of active analogs. The stimulation by active derivs. was concn.-dependent, and compds. could be ranked in potency according to their ED50 values for stimulation of AIB uptake: cortisol (I) [50-23-7] (44 nM) < prednisolone [50-24-8] (22 nM) < triamcinolone [124-94-7] (13 nM) < beclomethasone [4419-39-0] (7 nM) = betamethasone [378-44-9] (7 nM) < dichlorisone [7008-26-6] (4 nM) < dexamethasone [50-02-2] (2 nM). The order of potencies broadly parallels the relative daily oral replacement doses of these compds. in humans and indicates a high intrinsic potency of the 21-alc. chlorinated glucocorticoids, dichlorisone, and beclomethasone. .
