DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 14 The effects of bufadienolides (bufalin [465-21-4], bufotalin [471-95-4], cinobufagin [470-37-1], cinobufotalin [1108-68-5], gamabufotalin [465-11-2], and resibufogenin [465-39-4]) isolated from Senso (dried toad secretions) was compared to that of the cardenolides (digitoxin and ouabain) on cardiac function in a working guinea pig heart prepn. All the steroids had cardiotonic effects in a concn.Several reagents with their cas registry numbers 465-39-4 and 465-21-4 are used here.-dependent manner, and the min. threshold concn. was 10-8M for bufalin, cinobufagin, gamabufotalin, and digitoxin and 10-7M for bufotalin, cinobufotalin, and ouabain. In addn., the order of max. efficacy of cardiotonic action was as follows: cinobufagin (3 ′ 10-7M) = ouabain (3 ′ 10-7 M) > digitoxin (1 ′ 10-7 M) = gamabufotalin (3 ′ 10-7 M) > cinovufotalin (3 ′ 10-7 M) > bufotalin (1 ′ 10-7M). The effect of cinobufagin was examd. on exptl. induced heart failure caused by acute local ischemia by ligation of the left anterior descending coronary artery in the perfused guinea pig heart. Cinobufagin (3 ′ 10-7 M) and digitoxin (1 ′ 10-7 M) reestablished the coronary flow of perfused guinea pig heart to 90 and 98%, resp., of the level prior to the coronary artery ligation. The cardiac output and left ventricular pressure of perfused heart were increased to the level prior to the acute local ischemia, and the left ventricular work was increased by cinobufagin (3 ′ 10-7 M) and digitoxin (1 ′ 10-7 M) to 108 and 106%, resp., of the pre-ligation level. Thus, cinobufagin possesses strong cardiotonic action, similar, to digitoxin, in exptl. induced heart failure due to acute local ischemia. .
