DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Grayanotoxin I (G-I)(I) [4720-09-6], grayanotoxin II (G-II) [4678-44-8], and grayanotoxin III (G-III) [4678-45-9] were sepd. by a centrifugal partition chromatog. method using a solvent system of :40 CHCl3-MeOH-H2O (mobile phase, upper layer; stationary phase, lower layer) at a rotational speed of 700 rpm and a flow rate of 1 mL/min. The rotor of the centrifuge contained 12 column cartridges connected with each other in series. G-I, G-II, and G-III (5 mg each) were injected into the system. The constituents in the eluate were checked by TLC and quant. detd. at 460 nm after being treated with vanillin in a soln. of HClO4.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) A thin-layer chromatog. method was developed for the identification and quant. detn. of grayanotoxin I [4720-09-6], grayanotoxin II [4678-44-8], and grayanotoxin III [4678-45-9]. The optimum conditions were EtOAc-MeOH () as developing solvent, Merck Kieselgel 60 thin layer with 40% H2O, and 20% phosphotungstic acid in EtOH spray with 90° charring temp. for 20 min for spot detection. The spots were detected on a Chromatoscanner CS-920 at 492 nm. The calibration curves were linear for 0.5-2.5 mg/spot for std. samples with correlation coeffs. of 0.9987 for I, 0.9994 for II, and 0.9962 for III.
