Thus, a mixt. of quinizarin [81-64-1] 0.1, leucoquinizarin [476-60-8] 0.05, H3BO3 0.675, and 4-n-dodecylaniline [104-42-7] 0.45 mol in 75 g isobutanol [78-83-1] was heated at 105° for several h, cooled to room temp., dild. with 100 g xylene [1330-20-7], washed with several 100-g portions of H2O, and sepd. from the aq. layer to give 250 g blue soln. contg. 40 wt.% dye [42887-22-9]. Other dye solns. were similarly prepd. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Rat liver cytosol and buttermilk xanthine oxidase [9002-17-9] both converted 7-deoxypyrromycinone [21179-18-0], the 7-deoxyaglycone of marcellomycin, a new anthracycline antibiotic, to a nonfluorescent compd. under anaerobic conditions and in the presence of an electron donor. NADH and NADPH were equally effective electron donors for liver cytosol, and xanthine was the best cofactor for xanthine oxidase. However, xanthine was inactive with liver cytosol. 476-60-8 and 21179-18-0 which are cas registry numbers of chemicals are mentioned. Reactions with xanthine oxidase obeyed Menten-Michaelis kinetics and were inhibited by allopurinol. No xanthine oxidase activity was detected in liver cytosol. Xanthine oxidase also induced a loss of fluorescence when incubated with 7-deoxydaunorubicin aglycon [32384-98-8]. The nonfluorescent metabolite of 7-deoxypyrromycinone was tentatively identified as the dihydroquinonic deriv. of the parent deoxyaglycone [107806-66-6] on the basis of its spectrophotometric, fluorescent, thin layer chromatog., and mass spectral characteristics. Data demonstrate that more than one enzymic activity, xanthine oxidase, and an unidentified rat liver cytosolic enzyme convert the 7-deoxyaglycones of anthracycline antibiotics to nonfluorescent metabolites. .
