Biological actions of the natural coumarins: Biological actions of the natural coumarins. I. Changes of lipids and glucose levels in rat plasma after intraperitoneal administration of various linear-type furanocoumarins. Yasuda, Masahide; Baba, Kimiye; Kozawa, Mitsugi (Osaka Coll. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Expts. were carried out on the effect of various linear-type furanocoumarins on the concns. of lipids and glucose in rat plasma. It was found that bergapten (I) [484-20-8] and xanthotoxin [298-81-7] cause hypolipidemia. In contrast, imperatorin [482-44-0], isoimperatorin [482-45-1], oxypeucedanin (II) [26091-73-6] and oxypeucedanin hydrate [2643-85-8] cause hyperlipidemia. However, no effect on any lipid fraction in rat plasma was produced by isopimpinellin [482-27-9], phellopterin [2543-94-4], ferulin [29455-13-8], byakangelicin [482-25-7] or sec-O-acetylbyakangelicin [77196-05-5]. Glucose levels in rat plasma were increased by isopimpinellin and psoralen [66-97-7], and decreased by oxypeucedanin. These results suggest that the effects of linear-type furanocoumarin on lipid concn. in rat plasma depend upon whether substitution is present or not at the 5- and 8-posiiton of the skeleton and upon the kinds of substituents.

DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) Bergapten (I) [484-20-8], imperatorin (II) [482-44-0], and xanthotoxin (III) [298-81-7] were sepd. in Ammi majus ext. by TLC and detected by fluorometry. The ext. was chromatographed using silica gel G impregnated with 10% HCONH2 in MeOH as stationary phase and hexane-benzene-MeOH 7:2:1 as eluting system. The spots were outlined under UV light (254 nm) against stds. II and III can be detd. in a concn. range of 2-20 and 2-15 mg, resp., and I at 5-15 mg. The method was also applied to pharmaceutical tablets contg. 5 mg II and 10 mg III. The method is more rapid and highly sensitive compared to traditional methods.
