(HPLC) method, with liq.Several reagents such as 486-21-5 is used here.-liq. extn. and UV absorbance detection, has been developed for the quantification of Isofraxidin in rat plasma. The anal. was performed on a Diamonsil C18 column (200 mm ′ 4.6 mm i.d., 5-mm particle size) with acetonitrile-0.05% phosphoric acid, (vol./vol.), as isocratic mobile phase. The linear range was 0.05-8.0 mg mL-1 and the lower limit of quantification was 0.05 mg mL-1. The intra- and interday relative std. deviation (RSD) for measurement of 0.25, 2.0, and 6.0 mg mL-1 quality-control (QC) samples ranged from 5.7 to 6.4% and from 6.3 to 7.9%, resp. Accuracy, as relative error (RE), was from ±5.8% to ±7.3%. The method was validated for specificity, accuracy, and precision and was successfully used in a pharmacokinetic study of Isofraxidin in rat plasma after administration of Ciwujia ext. .

DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Factors (light, temp., humidity, etc.) affecting the contents of syringin [118-34-3], isofraxidin [486-21-5] and total flavones in the A. senticosus root or bulb during storage were detd. by TLC. Thesyringin and total flavone content decreased with storage time. Sunlight, temp. and humidity had little effect on the stability of isofraxidin, whereas the total flavone content was markedly lowered when the root or bulb was stored at higher relative humidity (74 or 86%) for 6 mo and syringin activity could not be detected when stored at 40-60° and 74-86% relative humidity or exposed to sunlight for 6 mon. The syringin activity in powd. roots or bulbs decreased to 50% of the original value after 12-mo storage and no activity was detected after 3-yr storage under regular conditions. .
