These sterols enhanced both sol. and particulate guanylate cyclase activities as well as cGMP [7665-99-8] levels 2-3-fold in human and rat tissues. At a concn. of 1 nM, vitamin D activities were 1,25(OH)2D3 [32222-06-3] > 25(OH)D3 [19356-17-3] > vitamin D3 [67-97-0] > 24,25(OH)2D3 [40013-87-4] = 25,26(OH)2D3 [29261-12-9] = vitamin D2 [50-14-6]. Dose-response curves revealed that maximal stimulation of guanylate cyclase by these sterols was at 1 nM and that there was no augmented guanylate cyclase activity at 0.01 nM. The precursors of vitamin D, cholesterol [57-88-5] and 7-dehydrocholesterol [434-16-2], had no effect on guanylate cyclase activity. The activation of guanylate cyclase activity by the vitamin D sterols required the presence of Mn2+. Ca2+ was not as efficient as Mn2+ in optimizing basal or hormone-stimulated guanylate cyclase activity. Vitamin D and its metabolites failed to stimulate adenylate cyclase (EC 4.6.1.1) [9012-42-4] activity. Thus, guanylate cyclase may play a role in the mechanism of action of vitamin D at the cellular level. .
