CODEN: BBIADT. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Lung strips of guinea pigs proved to be the most suitable in vitro model for the study of lipoxygenase [9029-60-1] inhibitors. Under basal conditions half-maximal contraction was produced by 13 mM arachidonic acid (AA) [506-32-1] which was strongly inhibited by 50 mM nordihyroguaiaretic acid (NDGA) [500-38-9] or 100 mM 3-tert-butyl-4-hydroxyanisole(BHA) [25013-16-5]. NDGA and BHA caused a shift of response towards higher concns. of AA. BHA also caused pronounced inhibitions of both basal tone and carbachol-induced contractions of the prepns. 506-32-1 and 25013-16-5 which are cas registry numbers of chemicals are mentioned. from the respiratory tract. Thus, the actions of NDGA and BHA on the airway prepns. are probably due to inhibition of the lipoxygenase reaction and, therefore, of leukotriene formation. The assumption of the lipoxygenase inhibition is supported by the action of the drugs on purified lipoxygenase from rabbit reticulocytes, which was inhibited 50% by 0.5 mM NDGA and 160 mM BHA. In contrast, the antioxidant 2,6-di-tert-butyl-4-hydroxytoluene [128-37-0] did not inhibit the enzyme at all, even at 1 mM. .

For example, TBHQ potentiated the antimicrobial activities of the preservatives against Staphylococcus aureus and Escherichia coli. Generally, the common order of increased activity regarding the effects of antioxidants on preservatives was TBHQ > PG > BHA > NDGA; TBHQ was the most sol. and NDGA was virtually insol. S. aureus, E. coli, And Candida albicans responded differently to the different antioxidants and preservatives. The Gram-pos. organism was inhibited by TBHQ but a varied effect was obsd. for Gram-neg. 1948-33-0 and 1138-60-9 are just another two chemicals used in this study. species. .
