DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) Serotonin creatinine sulfate (I) [971-74-4] and histamine-2HCl (II-2HCl) [56-92-8] produced dose-dependent contraction in bovine anterior, middle, posterior cerebral and basilar arteries. The sensitivity to I was greater than that to II in middle, posterior cerebral and basilar, but not in anterior cerebral, arteries. Only high concns. of l-norepinephrine bitartrate [51-40-1], methoxamine-HCl [61-16-5], and isoproterenol-HCl [51-30-9] caused a barely detectable contractile response in cerebral arteries. However, in the middle cerebral arteries, isoproterenol and norepinephrine (6 out of 10 strips) sometimes elicited a detectable relaxation which was blocked by propranolol. No response to acetylcholine chloride [60-31-1], carbachol [51-83-2], nicotine [54-11-5] or transmural stimulation was seen. Tyramine-HCl [60-19-5] (10-4M) only induced a small tonic contraction. Apparently, adrenergic and cholinergic receptors do not play a major role in regulating the muscular tone of bovine cerebral arteries. Regional differences of the cerebrovascular reactivity to biogenic amines were obsd.

Atropine blocked the cholinergic effect in both glands. The adrenergic discharge was inhibited by propranolol (a b-blocker) in parotid glands and by phentolamine (an a-blocker) in exorbital lacrimal glands. In the parotid gland, protein discharge evoked by norepinephrine was independent of the extracellular Ca2+, whereas in lacrimal glands, the adrenergic stimulation was almost lost in the absence of extracellular Ca2+. In both glands, the cholinergic stimulation of protein release was obsd. only in the presence of extracellular Ca2+. Another b-adrenergic agent l-isoproterenol d-bitartrate [54750-10-6] induced a large protein discharge in the parotid gland, but had no effect on lacrimal gland. Thus, protein output from both glands is independently induced by cholinergic and adrenergic agents through muscarinic receptors and 2 kinds of adrenergic receptors: b-adrenergic receptors for parotid glands and a-adrenergic receptors for the exorbital lacrimal glands.
