DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Glycogen [9005-79-2] deposition and glucose [50-99-7] tolerance were examd. in female mice after 24 days of oral treatment with natural (17b-estradiol [50-28-2] and progesterone [57-83-0]) and synthetic (ethinyl estradiol [57-63-6] and norethisterone acetate [51-98-9]) sex steroids, administered individually and in estrogen-progestin combination. Doses were 5 mg/kg/day for estrogens and 1 mg/kg/day for progestins. Compared with diestrus control mice, each treatment increased glycogen deposition in liver, uterus, heart, and biceps femoris muscle. 17b-Estradiol produced the greatest increments. Progesterone produced considerably smaller increments and antagonized the glycogenic effects of 17b-estradiol. Ethinyl estradiol and norethisterone acetate generally induced similar changes in glycogen deposition. Treatments with 17b-estradiol improved glucose tolerance. Although glucose tolerance was not significantly altered by the other sex steroid treatments, the changes in glycogen deposition indicate important effects on tissue carbohydrate metab.

Peak level, bioavailability and oral clearance of chloroquine were not significantly altered with the use of low dose combination pill. In another group of healthy women using the same contraceptive pills, serum levels of norethisterone (NET) [68-22-4] and EE were measured before and after 4 wk of chloroquine prophylaxis. There was no significant difference between peak level and AUC (0-24 h) of NET before and during chloroquine prophylaxis. There was a small (15%) but statistically significant increase in AUC (0-3 h) of EE during chloroquine prophylaxis. Ovulation inhibition was maintained during chloroquine therapy, as indicated by mild luteal serum progesterone levels below 2 ng/mL.
