DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 10 Arbekacin (HBK)(I) [51025-85-5] showed broad antibacterial spectra against gram-pos. and gram-neg. bacteria including Pseudomonas aeruginose. It was also effective against gentamycin [1403-66-3] or tobramycin [32986-56-4]-resistant bacteria. HBK was resistant to various aminoglycoside-inactivating enzymes except for AAC(2') [64060-44-2] and AAC(6')-IV [92479-98-6], both of which slowly inactivated it. Even at higher dosages (150 mg/kg i.m. or greater, which resulted in some details), HBK never decreased the pinna reflex in guinea pigs, while 150 mg/kg or more of dibekacin (DKB) or amikacin (AMK) caused loss of this reflex. HBK has less ototoxicity than do DKB and AMK. This was confirmed by histopathol. examn. of the inner ear. The degree of nephrotoxicity of HBK was similar to that of DKB as judged from serum biochem. tests, urinalysis, and histopathol.Except for chemicals metioned above, 56467-64-2 and 64060-44-2 are also used. findings. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) An enzyme immunoassay has been developed for the measurement of 4-amino-2-hydroxybutylyldibekacin (HBK)(I) [51025-85-5] a new semisynthetic aminoglycoside antibiotic. Antisera were raised in rabbits by immunization with HBK conjugated to bovine serum albumin (BSA). 3'-Eno-HBK conjugated to alk. phosphatase (ALP) was used as an enzyme-labeled antigen. The antibody-bound drug was sepd. from drug free using goat anti-rabbit IgG serum. The assay can be completed within 1 h by co-incubating the 1st and the 2nd antibody. The present immunoassay allows detection of 10 ng HBK per mL of serum, and is applicable for monitoring HBK level in blood. HBK concns. in human sera were detd. by the immunoassay during and after infusion and the levels were compared with those detd. by HPLC and antimicrobial assay.
