DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) D. rotundata flour (200 g) was hydrolyzed with 2N HCl and extd. with petroleum ether (40-60.degree.) to yield 0.6747 g of crude steroid. Purifn. afforded 0.1677 g of total sterols and 0.3021 g of sapogenins. D. cayenensis flour (200 g) similarly treated yielded 0.7841 of crude steroid, 0.2015 g of sterols, and 0.4 g of sapogenins. Further chromatog. and(or) mass spectroscopic anal. revealed that the sterols were .beta.-sitosterol [83-46-5], stigmasterol [83-48-7], campesterol [474-62-4], and cholesterol [57-88-5] and the sapogenins were diosgenin [512-04-9], yamogenin [512-06-1], kryptogenin [468-99-5], and gitogenin [511-96-6].

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) In mice the i.p. and intragastric LD50 values for diosgenin [512-04-9] were 3564 and >8000 mg/kg; in rats 4872 and >8000 mg/kg. In mice the i.p. and intragastric LD50 values for solasodin [126-17-0] were 898.7 and >8000 mg/kg; in rats 395.7 and 4978.5 mg/kg. On repeated administration of similar doses of these 2 compds. (250 mg/kg) diosgenin had no toxic effects, whereas solasodin induced toxic changes in the liver, kidneys, nervous system, and endocrine glands. The MPL for solasodin is 0.03 mg/m3.
