DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Rat granulosa cells were preincubated with FSH [9002-68-0] (1 mg/mL) and/or androgens (testosterone [58-22-0] and 5a-dihydrotestosterone [521-18-6], 0.5 mM) and subsequent 14C-labeled progesterone [57-83-0] metab. by these cells was studied. Granulosa cells metabolized radiolabeled progesterone to 3 major products, 20a-hydroxy-4-pregnen-3-one [145-14-2], 3a-hydroxy-5a-pregnan-20-one [516-54-1], and 5a-pregnane-3a,20a-diol [566-58-5]. Androgens, but not FSH, decreased overall progesterone utilization. Both FSH and androgens decreased conversion of progesterone to 20a-hydroxy-4-pregnen-3-one. The accumulation of 5a-pregnane-3a,20a-diol was increased by FSH, decreased by testosterone, and not altered by 5a-dihydrotestosterone. The accumulation of 3a-hydroxy-5a-pregnan-20-one was increased by both FSH and androgens. Apparently, FSH as well as androgens modulate progesterone metab. by rat granulosa cells. FSH appears to increase 5a-reductase [9036-43-5] activity and/or decrease 20a-hydroxy steroid dehydrogenase [9040-08-8] activity, whereas androgens decrease only 20a-hydroxy steroid dehydrogenase activity. .

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The effects of androgens on progesterone [57-83-0] accumulation, utilization of exogenous progesterone, and accumulation of [4-14C]progesteroine metabolites by rat granulosa cells in culture were studied. Androgen increased progesterone accumulation in cultures without exogenous progesterone and slowed the overall decline of progesterone concn. in cultures supplemented with exogenous progesterone. Both aromatizable testosterone [58-22-0] and nonaromatizable 5a-dihydrotestosterone [521-18-6] decreased [4-14C]progesterone utilization by granulosa cells by 12-30%. Androgens decreased conversion of radiolabeled progesterone to 20a-hydroxy-4-pregnen-3-one [145-14-2] by 11-50% and to 5a-pregnane-3a,20a-diol [566-58-5] by 26-49%. Accumulation of 3a-hydroxy-5a-pregnan-20-one [516-54-1] was not altered in 3 h incubations and was increased by up to 43% in 24 h incubations by androgen treatment. Androgens apparently alter progesterone catabolism by granulosa cells by decreasing 20a-hydroxy steroid dehydrogenase [9040-08-8] activity and this effect may contribute to the overall stimulatory action of androgens on progesterone accumulation.
