DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) A sensitive and chem. specific high-speed liq. chromatog. method was developed for the detn. of 4-nitro-2-phenoxymethanesulfonanilide (I) [51803-78-2] in plasma. The method includes selective extn. of the antiinflammatory drug and an internal std., 2-(4'-chlorophenoxy)-4-nitromethanesulfonanilide, into benzene from acidified plasma followed by reextn. into 0.2 N NaOH. The aq. layer is acidified, and the drug is reextd. into benzene. The benzene is evapd., and the residue is dissolved in a small vol. of acetonitrile. At 10-mL aliquot is analyzed on a reversed-phase column. A m Bondapak C18 reversed-phase column is used with 50% acetonitrile in distd. water as the elution soln. A fixed-wavelength (UV) detector (254 nm) is used. The mean overall extn. recovery, after correction for aliquot factors, is 99%. The accuracy, expressed as the relative error, is 4, 0.3, and -3% at 0.60, 1.50, and 3.00 mg/mL, resp. Repeated anal. of ref. stds. indicates that the precision, expressed as the relative std. deviation, is 3% or less. The lower sensitivity limit is 0.2 mg/mL with a 2-mL plasma sample. The method was applied successfully to the detn. of plasma levels of I in humans and rats in metabolic expts. at pharmacol. doses.
