DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) In dogs, neurotensin [39379-15-2] (10 .mu.g/kg) injected into the pancreaticoduodenal artery caused a marked biphasic increase blood plasma insulin [9004-10-8] and proinsulin C-peptide [59112-80-0]. Plasma glucagon [9007-92-5] increased approx. 3-fold, plasma gastrin [9002-76-0] increased over 3 times, and plasma glucose increased about 17%. Xenopsin [51827-01-1] (10 .mu.g/kg) resulted in a smaller biphasic increase of insulin compared to neurotensin, but a similar response in glucagon secretion, accompanied by a rise in glucose of approx. 15%. Plasma gastrin increased up to 3 times. .

DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) The effects of neurotensin [39379-15-2] on plasma insulin [9004-10-8] and gastrin [9002-76-0] secretion were investigated in 12 normal and 8 hypophysectomized dogs. Neurotensin was injected rapidly or infused for 30 min into the superior pancreaticoduodenal artery. In normal dogs, rapid administration of neurotensin (10 .mu.g/kg) brought about a mild hyperglycemia and a rapid and sharp increase of plasma insulin and gastrin levels. In the pancreatic vein, a biphasic insulin response was noted. The same secretory pattern of insulin was demonstrated during neurotensin (500 ng/kg/min) infusion. In the antral vein, the plasma gastrin level increased rapidly to 425% from basal level within 2 min and returned nearly to the initial level within 10 min after a rapid administration of neurotensin. In hypophysectomized dogs, the basal levels of plasma glucose, pancreatic insulin and antral gastrin were decreased to 53%, 9%, and 65% of normal dogs, resp. Neurotensin had little effect on insulin secretion, and a biphasic insulin secretory pattern obsd. in normal dogs was not demonstratedin hypophysectomized dogs. However, neurotensin-stimulated plasma gastrin concns. were considerably maintained. Also, xenopsin [51827-01-1] (10 .mu.g/kg) induced a milk hyperglycemia, hyperinsulinemia,and hypergastrinemia, but its secretory activities were smaller than those of neurotensin. Neurotensin apparently stimulates insulin and gastrin secretion by neurogenic vasodilatation or by a direct acion an pancreatic .beta.-cell and gastrin secretory cell of the gastric antrum.
