DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) DL-.alpha.-tocopheryl nicotinate (I) [51898-34-1] (0.04 or 0.2 mmol/day/kg orally) or DL-.alpha.-tocopheryl acetate (II) [7695-91-2] (0.2 mmol/day/kg orally) delayed the progress of hypertension in unilaterally nephrectomized rats treated with deoxycorticosterone and salt, and in young genetically hypertensive rats (SHR) given NaCl soln. Treatment with I or II reduced the suppression of body wt. gain, incidence of pneumonia, and mortality in hypertensive rats. Severe hypertension in old SHR (9 months) progressed further when the rats were given NaCl soln. instead of drinking water, but treatment with I or II prevented the increase in blood pressure and the incidence of stroke.

DOCUMENT TYPE: Journal CA Section: 18 (Animal Nutrition) The metab. of the nicotinate (I) [59-67-6] moiety of dl-a-tocopherylnicotinate (II) [51898-34-1] in rats was compared with that of free I. When a I-free, tryptophan [73-22-3]-limited diet contg. 0.065% II was fed to weanling rats for 14 days, they grew as well as rats fed a tryptophan-limited diet contg. 0.015% I and 0.05%-dl-a-tocopherol. After 15 days, the I-free, tryptophan-limited diet was fed to both groups of rats. 73-22-3 and 3106-60-3 which are cas registry numbers are also used here. As a result, body wt. gains of rats previously fed the II diet were superior to those of rats previously fed the I diet. Urinary excretion of niacin [59-67-6] and N1-methylnicotinamide [3106-60-3] was lower in the II group than in the I group. Evidently, body retention of II is higher than of I. Even when a 20% casein diet contg. large amt. of II (0.5%) was fed to weanling rats, wt. gains were almost the same as of rats fed the 20% casein diet and the 20% casein diet contg. 0.11% I and 0.39% dl-a-tocopherol. The NAD [53-84-9] levels of liver, kidney, and blood in the II group were higher than in the group fed 20% casein, but lower than in the I group. Urinary excretion of niacin, N1-methylnicotinamide, N1-methyl-2-pyridone-5-carboxamide [701-44-0], and nicotinuric acid [583-08-4] were higher on the I and II diets than on the 20% casein diet, but with II causing smaller increases. Apparently the turnover rate of the I moiety of II is slower than that of free I. .
