DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) Optimal conditions for the detn. of emodin (I) [518-82-1] and physcion [521-61-9] in the bulb of P. cillinerve by pulse polarog. were established. A 0.5-g sample was extd. with MeOH and the ext. was chromatographed on a silica gel G plate by using hexane-ether-EtOAc-CH2Cl2-formic acid (8:1.5:1:0.8:0.1) as eluent. Spots corresponding to free I and physion were scrapped, extd. with EtOH and an aliquot (1.0-4.0 mL) with addn. of EtOH, 4.0 mL LiCl (0.1M) and 1 drop of dil. NH4OH was analyzed by pulse polarog. The current was linearly correlated to the concn. in the range of 6.4 ′ 10-6-3.1 ′ 10-5 and the relative std. deviation was >2%. For the detn. of conjugates, samples were hydrolyzed with HCl to give total I and total physcion. Based on these results (e.g. total I anthraquinones-free anthraquinones), conjugated anthraquinones were calcd. Thus, the bulk of P. cillinerve contained 0.05% free I, 3.10% free physcion, 0.4% I conjugate and 9.80% physcion conjugate.
