DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The behavioral actions of tetrahydroberberine (I) [522-97-4] were compared to those of tetrahydrocoptisine (II) [7461-02-1], tetrahydropalmatine (III) [10097-84-4], tetrahydrojatrorrhizine (IV) [13063-54-2], chlorpromazine and chlordiazepoxide. I had anti-mescaline, anti-apomorphine, anti-methamphetamine activities and inhibited conditioned avoidance responses. I influenced the electroencephalog., esp. the enhancement of after-discharge in limbic system as did chlorpromazine. I did not cause catalepsy, but enhanced that induced by chlorpromazine. On the other hand, the acute toxicity of I was less than that of chlorpromazine and chlordiazepoxide. In its effects on the nervous system, I was equipotent to II and III, but more potent than IV. I did not cause hibernation, muscle relaxation, or disappearance of righting reflex. Thus I may be useful as a tranquillizer.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Tetrahydroberberine (I) [522-97-4] and l-tetrahydropalmitine (II) [483-14-7] antagonized apomorphine- and amphetamine-induced turning behavior in rats with 6-hydroxydopamine-induced lesions of the nigrostriatal dopaminergic pathway, thereby showing a dopamine-receptor-antagonistic activity similar to that of haloperidol. l-Stepholidine (III) [16562-13-3] antagonized the turning response to amphetamine but potentiated that to apomorphine, suggesting that III also possesses partial agonist properties. III was 3-5-fold more potent than I or II. The effects of I could be reversed by scopolamine, suggesting the involvement of neostriatal muscarinic receptors in the blockade of postsynaptic dopaminergic receptors by I. From these and other data, it is suggested that these 3 tetrahydroprotoberberine analogs form a new chem. class of brain dopaminergic receptor antagonists.
