DOCUMENT TYPE: Conference CA Section: 4 (Toxicology) There was a strong assocn. between arom. hydrocarbon responsiveness and N-hydroxylation of phenacetin [53-96-3], acetanilide [103-84-4], and p-chloroacetanilide [539-03-7] in livers from progeny and the appropriate backcrosses and intercross involving B6 and D2 mice. Mutagenic activity in the Salmonella test system of several 2-acylaminofluorenes were compared using the S-9 liver fractionsfrom B6 and D2 mice. Pretreatment of B6 mice with 3-methylcholanthrene(3-MC) increased the mutagenicactivity of all the analogs in the S-9 liver fraction. No increase in mutagenic activity was found in S-9 liver fraction from D2 mice after 3-MC pretreatment, indicating that mutagenic activity correlates with arom. hydrocarbon responsiveness and N-hydroxylase activity. a-Naphthoflavone, an inhibitor of N-hydroxylation of 2-acetylaminofluorene (I) [53-96-3]in vitro, inhibited the mutagenic activity of these analogs indicating that N-hydroxylation is the 1st step in the activationof N-acylaminofluorenes into mutagens.

(GLC) coupled with column chromatog. was used to accurately det. as little as 25 ppm p-chloroacetanilide (I) [539-03-7] in acetaminophen (II) [103-90-2]. I was eluted from a pH 8 phosphate-buffered diatomite partition column by using purified tetrachlorethylene (II was retained). This soln. was concd., internal std. (docosane) was added, and I was detd. by using a 0.9 m ′ 2 mm glass column packed with 3% Poly A 103 on Supelcoport and a flame ionization detector with electronic integration. Std. curves were linear for 10-100 ppm I. Various chromatogrphic materials were investigated for optimal retention characteristics. High pressure liq. chromatog. (HPLC) was also evaluated as an alternative; however, lack of reproducitiblity of the HPLC column favored the GLC procedure.
