DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The antihistaminic effects of 2-guanidinebenzimidazole (GBI)(I) [5418-95-1] and 1-phenylbiguanide (PBB)(II) [102-02-3] on betazole stimulated gastric acid secretion were confirmed by the inhibitory activity of these compds. on the isolated guinea pig auricle stimulated by histamine. The antimuscarinic activity of GBI and PBG on carbacholstimulated gastric acid secretion in rats is supported by the way in which these same drugs depress the motility of the duodenum and colon of the anesthetized cat stimulated by prostigmine. The above mentioned effects of these compds. are also assocd. with myolytic activity, since they decrease the spontaneous with myolytic activity, since they decrease the spontaneous and histamine-stimulated motility of the duodenum and colon. GBI and PBG probably depress gastric acid secretion by interfering with both histamine and acetylcholine receptors and with other sites involved in the secretory process. GBI and PBG appear to be capable of decreasing gastric acid secretion, while the compds. dimethylbiguanide [657-24-9] and cyanoguanidine [461-58-5] do not. Thus, the antisecretory effect is present when the biguanide groups are assocd. with lipophilic mols.

Thus, 2-benzimidazolylguanidine [5418-95-1] was alkylated with chlorinated polyisobutylene (mol. wt. 960; 4 wt.% Cl) at 200.degree. for 4 h. The product (contg. 1.4 wt.% N and 0.2 wt.% Cl), added to a lubricating oil at 2 wt.%, had better dispersant properties than conventional alkylsuccinimide dispersants, and showed good detergent characteristics in the Petter AVI engine test. Similarly alkylated and used were 2-benzothiazolylguanidine [2582-07-2], 2-benzoxazolylguanidine [39123-82-5], and 2-(4(3H)-oxoquinazolinyl)guanidine [62936-92-9]. The same dispersants were also effective in gasolines at 200 ppm.
