Selective extraction of DNA from fixed mammalian tissues with formic acid
Selective extraction of DNA from fixed mammalian tissues with formic acid. Dutt, Mihir K. (Dep. Zool., Univ. Delhi, Delhi, India). Indian J. Exp. Biol., 15(12), 1219-20 (English) 1977. CODEN: IJEBA6. ISSN: 0019-5189. DOCUMENT TYPE: Journal CA Section: 9 (Biochemical Methods) Differential extn. of DNA from fixed mammalian tissue sections is possible by treatment with boiling 20 and 25% HCO2H for 25 min, as judged by Methyl green and Pyronine G as well as with the regular Feulgen procedures. Treatment of sections with HCO2H at 60° for 25 min caused depolymn. of DNA as indicated by the red nuclei with Pyronine G staining.
Methyl green-pyronin Y staining of nucleic acids: studies on the effects of staining time, dye composition and diffusion rates
Methyl green-pyronin Y staining of nucleic acids: studies on the effects of staining time, dye composition and diffusion rates. Prento, P.; Lyon, H. O. (Department of Pathology 134, Hvidovre Hospital, University of Copenhagen, Hvidovre DK-2650, Den.). Biotechnic & Histochemistry, 78(1), 27-33 (English) 2003 BIOS Scientific Publishers Ltd. CODEN: BIHIEU. ISSN: 1052-0295. DOCUMENT TYPE: Journal CA Section: 9 (Biochemical Methods) Section cross-reference(s): 3, 13 Since the introduction of the methyl green-pyronin Y procedure as a differential histol. stain more than 100 yr ago, the method has become a histochem. procedure for differential demonstration of DNA and RNA. Numerous variants of the procedure have been suggested, and a no. of hypotheses have been put forward concerning kinetics and binding mechanisms. Using both filter paper models contg. DNA, RNA or heparin and histol. sections, we have attempted to evaluate the kinetics of staining and the role of staining time for methyl green and pyronin Y by applying the dyes individually, simultaneously and sequentially. The results are presented as color charts approximating the obsd. staining patterns using a computerized palette. Our results indicate unequivocally that the differential staining is not time-dependent, but that it is dictated by the relative concns. of methyl green and pyronin Y and by the pH of the staining soln.
