DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Subjects using 30 mM Zn phenolsulfonate [127-82-2] as a mouthwash retained 12% of the Zn. Salivary Zn concn. was increased by using mouthwashes contg. 17-35 mM Zn as ZnSO4, Zn phenolsulfonate, or Zn citrate [546-46-3]. For 17 mM ZnSO4 or Zn phenolsulfonate, the effect lasted 3-4 h. Zn retained in the mouth gave visible fluorescence after rinsing with 8-hydroxyquinoline, and this was particularly evident on the tongue, cheek mucosa, and dental plaque. The concn. of Zn in plaque was increased 13-19-fold 1 h after using 31 or 18 mM Zn phenolsulfonate. A 3-fold increase was still present 6 h later for the 31 mM mouthwash. Zn salts inhibited acid prodn. from [14C]glucose in vitro by plaque at concns. which were found in plaque in vitro after using the mouthwashes. The effect of Zn on the metabolic activity of plaque may reduce the growth rate of plaque bacteria and so decrease plaque growth. .

The effectiveness of the treatment was reduced by increasing the time allowed for growth after the final application of Zn. Zn citrate [546-46-3] and Zn phenolsulfonate [127-82-2], 17-9 mM, resulted in about a 30% redn. in the extension of plaque along the gingival margin in vivo over 16 h. Subjects with high rates of plaque growth with the placebo mouthwash were most affected by Zn salts. There are some commonly used reagents like 546-46-3 in this article. Increasing the Zn concn. to 30 mM did not further increase the activity. The low activity of Zn against plaque growth in vivo may be due to a low concn. of Zn or a low frequency of application. Thus, Zn salts, including Zn citrate inhibit the growth of plaque in vivo and in vitro. .
