Other cephalosporins, such as cefotaxime [63527-52-6], were much less active. In spite of their relatively good in vitro activities, however, neither cefazedone nor cephalotin reduced the bacterial counts in mice infected with a virulent strain of L. monocytogenes. In contrast, ampicillin [69-53-4] was active both in vitro and in vivo. Cephalosporins are not effective in treatment of listeriosis.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Among ten cephalosporins injected i.m. into rats at 150 mg/kg, cefazolin [25953-19-9], cefazedone [56187-47-4], and cefuroxime (I) [55268-75-2] gave large areas under the serum concn.-time curve, whereas cephalothin [153-61-7], cephapirin [21593-23-7], and cephradine [38821-53-3] gave small areas; other compds. were intermediate. I and cefamandole [34444-01-4] (150 mg/kg i.m. twice a day for 7 days) were most effective against both estrogen-induced and non-estrogen-induced Escherichia coli pyelonephritis in rats; cephapirin and cephradine were less effective. Cefazolin, which was relatively ineffective against estrogen-induced pyelonephritis, was as effective as I and cefamandole against non-estrogen-induced pyelonephritis. The therapeutic efficacy of the drugs was to some extent related to their pharmacokinetics and serum protein binding.
