DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Mutagenic activities of 4-aminopyridine (4AP) (I) [504-24-5], 4-aminoquinoline (4AQ) [578-68-7], 9-aminoacridine (9AA) [90-45-9] and harman (HM) [486-84-0] were examd. by the Salmonella test system in the presence of CoCl2, which itself is nonmutagenic in this system. Mutagenic activity of the mixt. of 9AA and CoCl2 was much higher than that of 9AA alone in strains TA1537 and TA2637. A similar enhancing phenomenon was obsd. in 4AQ-CoCl2 and HM-CoCl2 mixts.There are some commonly used reagents with their cas registry numbers 5893-73-2 and 486-84-0 in this article. but not in that of 4AP-CoCl2. Judging from visible and NMR spectral data, this increased mutagenicity may be attributable to the formation of moderate to weak complexes between these chem. and the Co2+. A survey of the mutagenicity of several Co2+ complexes supported this interpretation. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effects of 4-aminoquinoline (4-AQ) (I) [578-68-7], a drug structurally related to 4-aminopyridine, were studied on atrial contractile fibers from the amphibian Caudiverbera caudiverbera by means of intracellular electrodes. In concns. that did not change resting potential, 4-AQ (0.25-1.5 mM) reduced upstroke velocity, action potential amplitude (APA), and overshoot in a concn.-dependent manner. 4-AQ also prolonged action potential duration (APD) and the fiber effective refractory period (ERP), making the ratio ERP/APD >1. These effects were reversible by washing. The drug-induced redn. of upstroke velocity was frequency-dependent. This effect and increase of APD were more intense at higher values of resting potential. All effects produced by 4-AQ remained const. in either high or low external K+ concns. The depressant effects of this compd. on upstroke velocity, overshoot, and APA as well as the increase of APD were antagonized by doubling the extracellular Na+ concn. The data support the view that 4-AQ exerts its effects on the frog heart by blocking both Na+ and K+ channels, thus causing quinidine-like actions.
