59. Inhibition by poly(vinylpyrrolidone) (PVP) of gastrointestinal absorption of salicylic acid and related compounds in rats and its mechanism from the viewpoint of intermolecular interaction. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Poly(vinylpyrrolidone) (PVP) [9003-39-8] inhibited gastrointestinal absorption of salicylic acid (I) [69-72-7] and related compds.Some commonly used reagents like 65-85-0 and 69-72-7 are used in this experiment. in rats in the decreasing order of 2,4-dihydroxybenzoic acid [89-86-1], p-aminosalicylic acid [65-49-6], salicylic acid, salicylamide [65-45-2], and o-anisic acid [579-75-9]. This order of inhibition was independent of the easiness of absorption of these compds. in the absence of PVP. The inhibitory effect of PVP on the absorption was obsd. to be reflected on the blood level pattern of salicyclic acid administered orally. Intermol. interaction of 12 compds. related to salicylic acid with PVP was analyzed by the equil. dialysis method. The degree of the interaction, expressed in terms of R-value, ran parallel with the degree of inhibition of the gastrointestinal absorption by PVP. From the relationship between the chem. structure and R, it was presumed that hydrogen bonding through the phenolic hydroxyl group of the small-mol. compds. played an important role in the interaction with PV. Such a presumption was supported by the results of IR spectroscopic examns. .
