DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The effect of sheep plasma erythropoietin [11096-26-7] prepns. on the incorporation of 3H-labeled uridine [58-96-8] into erythroid cells was studied using cells of fetal calf liver cultured in serum-free medium. The cells were incubated for 20 h with the hormone, followed by a 1-h incubation with [3H]uridine. Erythropoietin caused a 2.5-fold increase in the incorporation of uridine into cold TCA-insol. cell exts. and a 70% increase in the incorporation of uridine into the cold acid-sol. cell exts. The phosphorylated metabolites of labeled uridine present in the cold acid-sol. fraction were analyzed by anion-exchange high HPLC. Erythropoietin increased the amts. of labeled UDP [58-98-0] and UTP [63-39-8] per cell. However, the specific activity of UTP and the labeled amts. 11096-26-7 and 58-96-8 which are cas registry numbers of chemicals are mentioned. of UDP-glucose in erythropoietin-treated cells were not different from those in control cell cultures. After chromatog. of the crude erythropoietin prepns. on reversed-phase and gel-permeation HPLC, there was a perfect coincidence of the fractions stimulating uridine incorporation into acid-sol. and acid-insol. cell exts. The protein fractions from crude erythropoietin which stimulated uridine incorporation after purifn. by reversed-phased HPLC were also able to stimulate globin chain synthesis in fetal calf liver cells. Apparently, the multiple effects on uridine metab. described above are due to erythropoietin, rather than other proteins contaminating the crude hormone prepns. .

UDP pretreatment accelerated blood EtOH disappearance rate by 50%. Increases of the hepatic triglycerides 6 h and 20 h after EtOH administration were lowered in UDP-pretreated rats. Hepatic ATP [56-65-5] contents and the size of the adenylate [61-19-8] pool increased in the UDP-pretreated group. Thus, UDP enhances EtOH metab. possibly by causing a change in ATP metab.
