DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The mutagenicities of 6 monocyclic aroma amines (2,4-diaminoanisole (I) [615-05-4], 2,4-diaminoethoxybenzene [5862-77-1], 2,4-diaminopropoxybenzene [37878-54-9], m-phenylenediamine [108-45-2], 2,4-diaminotoluene [95-80-7], and nitro-p-phenylenediamine [5307-14-2] were investigated in S. typhimurium strains TA 97, 1537, and 1538 in the presence of 2 different amts. of S9 prepns. Strain TA1538 was the most responsive of the 3 strains with this group of compds. TA 1537 was more responsive to 3 of the compds. than strain TA 97, in terms of the mutagenicity in proportion to background. Regarding the no. of revertants per nmol or the no. of induced revertants per plate, TA 97 was more responsive than TA 1537 for all 6 compds. Comparisons of mutagenesis from tests involving strain TA 97 are complicated by the large variations in spontaneous mutation frequency in this strain. The amt. of S9 per plate is another important variable in tests of monocyclic arom. amines. In general, more revertants were detected with 10% S9 than with 4% S9. For 2,4-diaminoanisole, 2,4-diaminoethoxybenzene, and 2,4-diaminopropoxybenzene, the same relation between chem. structure and mutagenic activity was obsd. In all 3 strains, a lower mutagenic response corresponded to a larger alkoxy substituent on the test mol.
