DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The thiobarbituric acid (TBA)-reactants were measured in the circulating red blood cells (RBC) and RBC trapped in the spleens of normal and phenylhydrazine-HCl [59-88-1]-treated rats. There was significant TBA-reactivity in the circulating RBC of phenyl-hydrzine-treated rats which was increased 3-fold in RBC obtained from the spleen. Since lipid peroxidn. accompanies formation of TBA-reactive malonyldialdehyde, it is suggested that phenylhydrazine induced anemia as a consequence of peroxidn. of RBC membrane lipids and this effect may be a result of the autoxoidn. of the drug and the interaction of O radicals with membrane lipids.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The effect of phenylhydrazine hydrochloride (PHH) [59-88-1] administration on the lipid compn. of spleen and liver was investigated in rats. There was a progressive splenomegaly with no change in liver size. There was an increase in the triglyceride content of liver in the treated animals which contributed to the increased total lipid content. The spleen had a decrease in total lipid content due to redn. in neutral lipids. The phospholipid level of both spleen and liver was not altered. Among the phospholipids of liver, the proportion of phosphatidylethanolamine increased while that of phosphatidylcholine decreased. Histol. studies showed degeneration of hepatic cells with fatty change. It appears that lipids of liver and spleen do not contribute to the hemolytic process operating in PHH-treated animals.
