(tyramine [51-67-2], hydroxylamine [7803-49-8], hydrazine [302-01-2], DMSO [67-68-5], KNCO [590-28-3], and acrylamide [79-06-1]). Mutagenicity of the chem. compds. and g-rays was examd. in Chinese hamster cell culture in the 1st and 2nd mitotic cycle by both MT and chromosomal aberrations. The no. of cells with the micronuclei in the 2nd mitotic cycle corresponded with the no. of aberrant cells in the 2nd cycle following g-irradn. The av. no. of aberrant cells was ~2-fold normal. In the 2nd cycle, not only the cells with the micronuclei increased under the effect of chem. agents [caffeine (I) [58-08-2]], but pyknotic cells also increased (which reduces the percentage of cells with micronuclei). Thus, while estg. mutagenicity of chem. compds. both cells with micronuclei and pyknotic cells should be considered.

The cells were treated with mutagens and then thermally treated at once (42°, 15 min; instantaneous induction) or incubated at 32° for 90 min and then thermally treated (delayed induction). N-Nitroso-N-methylurea (NMM) [684-93-5] had the strongest mutagenic and lethal action and was studied in detail. In a delayed induction, the survivability of the prophage increased and the frequency of c mutations decreased irresp. of the repair genotype of the host. Mutagenic effects of NMM were not affected by carbamoylation since 0.2M KNCO had almost no inactivating effect on the prophage and did not induce any mutation. MNNG [70-25-7], with a similar mechanism of action to NMM, gave qual. similar data. Et methanesulfonate (EMS) [62-50-0] was less mutagenic than MNNG and NMM. EMS induces mutations by a replicative mechanism and the max. yield does not depend on recA+. Me methanesulfonate [66-27-3] induces prophage c mutations in recA+ much better than in recA-; it is a repair-type mutagen. Nitrogen mustard [55-86-7] causes a rapid inactivation of the prophage (recA-) but does not induce c mutations. Mutation yields with mitomycin c [50-07-7] and acridine mustard [146-59-8] were small. The last 2 bifunctional alkylating agents presumably exert their mutagenic action via intercalation.
