(I): R = H,Me; R1 = H, Me, cyclopropylmethyl, cyclobutylmethyl, cyclopropylethyl, allyl) were prepd. from 6-hydroxytropinone [5932-53-6] by ketalization, Pfitzner-Moffit oxidn., Grignard reaction with p-methoxyphenyl bromide [104-92-7], dehydration, hydrogenation, treatment with methylidene or ethylidene triphenyphosphorane, and cyclization with HBr, followed by N-demethylation, N-acylation, and redn. All the compds. were fairly toxic in relation to their weak analgesic properties. I(R = H, R1 = cyclopropylmethyl) [61727-63-7] and I(R = Me, R1 = cyclopropylmethyl) [61727-64-8] had narcotic antagonist properties considerably greater than pentazocine and approx. equiv. to nalorphine. Structure-activity relations and the correlation of analgesic and narcotic antagonist properties in series of compd. derivs. are discussed. .
