Synthesis of a 19-residue peptide with alamethicin-like activity
Synthesis of a 19-residue peptide with alamethicin-like activity. Gisin, Balthasar F.; Kobayashi, Shigeru; Hall, James E. (Dep. Biochem., Rockefeller Univ., New York, N. Y., USA). Proc. Natl. Acad. Sci. U. S. A., 74(1), 115-19 (English) 1977. CODEN: PNASA6. DOCUMENT TYPE: Journal CA Section: 34 (Synthesis of Amino Acids, Peptides, and Proteins) A compd. with voltage-gating characteristics similar to those obsd. in nerve membranes was prepd. The open chain sequence of alamethicin (ALA-o) proposed by Martin and Williams (1976) was synthesized by stepwise solid-phase condensation of 4 fragments and was not identical with the main component of natural ALA. In lipid bilayer membranes the exponential dependence of cond. on voltage and the dependence of cond. on a high power of the peptide concn. were qual. similar for ALA-o and for natural ALA.In this experiment, several chemicals are used like 59588-86-2 and 62571-14-6 ALA-o showed the characteristics of a channel-former, although the single-channel conductances were less well defined for the synthetic compd. A cyclic structure is not necessary for a peptide to induce voltage-depended conds. in membranes. .
