DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) Inanesthetized cats, self-stimulation with systematically varied stimulus voltages was tested under the effect of acetylcholine chloride [60-31-1], adrenaline-HCl (I) [55-31-2], noradrenaline [51-41-2], and vasopressin [11000-17-2]. Heart rate, respiration, and arterial blood pressure were recorded simultaneously with the lever-pressing rate. Both facilitation and suppression of self-stimulation was induced by all of these drugs, depending on the dosage and on the stimulus voltage applied. Conspicuous changes in peripheral autonomic functions appeared regularly with alterations in self-stimulation behavior. The facilitation or suppression of self-stimulation seemed to be related to the magnitude of the autonomic changes caused by the drugs. Whenever the changes in the peripheral autonomic functions decreased below, or increased above a certain level, the animal stopped self-stimulating. Within these levels the relation between the amplitude of drug effects and that of original effects of self-stimulation seemed to be crit. for an alteration of self-stimulation. Whenever the autonomic effects of a drug overreached the autonomic effects of control self-stimulation, the lever-pressing rate decreased. In contrast, if the original effects of self-stimulation exceeded the effects of the drug, then the lever-pressing rate increased. These drugs may have a nonspecific effect on self-stimulation behavior. Apparently, the peripheral regulatory mechanisms need to be incorporated into the present theories of drug effects on self-stimulation behavior.

On the other hand, there was a remarkable increase in th bronchodilator effect of isoproterenol-HCl [51-30-9] and aminophylline [317-34-0]. Thus, in the asthmatic patients, I may act directly on the bronchial smooth muscle, potentiating the bronchodilator effect of catecholamines and b-adrenergic stimulating drugs and attenuating the bronchoconstrictor effect of various bronchoconstrictors.
